No. A DMCA notice can be sent based on unregistered copyright. Registration becomes essential if you want to sue, and it makes a claim far harder for a platform or infringer to brush off.
That is a trademark complaint rather than a DMCA notice, and it goes through a different platform process. Platforms generally require a registration number for trademark claims, which is one more reason to register.
